Induced pluripotent stem cell (iPSC) technology may support genetic rescue; however, iPSC derivation in Bovidae remains challenging. Here, we investigated whether bovine preimplantation embryos provide a permissive environment for wisent somatic cells expressing reprogramming factors. Wisent adult fibroblasts were engineered to express either the fluorescent reporter mKO2 alone (BAF-KO2) or mKO2 together with six reprogramming factors (OCT4, SOX2, KLF4, c-MYC, NANOG, and LIN28; BAF-6TF-KO2) delivered using the PiggyBac transposon system. Five wisent cells were microinjected into eight-cell-stage bovine embryos produced by in vitro fertilization. A subset of BAF-6TF-KO2 cells was additionally cultured under pluripotency-promoting conditions before injection (BAF-6TF-KO2-IND). Injected embryos developed to the blastocyst stage at rates comparable to sham-injected controls. mKO2-positive wisent cells were detected in 75% of blastocysts injected with BAF-KO2 and 91% of those injected with BAF-6TF-KO2 (p < 0.05). Cells expressing reprogramming factors localized within the embryo significantly more often than controls. Most wisent cells showed limited proliferation and signs of compromised nuclear integrity. However, a subset of BAF-6TF-KO2 cells expressed CDX2, suggesting responsiveness to trophectoderm-specifying signals. These findings show that bovine preimplantation embryos can accommodate wisent somatic cells up to the blastocyst stage and suggest that reprogramming-factor expression improves their retention and internal localization. This study provides a foundation for further development of interspecies embryo complementation approaches in Bovidae for conservation purposes.","interpretation_basis":null,"fulltext_status":null,"interpretation":null,"fulltext":{"available":false,"status":null,"reason":"尚未抓取全文（未入选、分数未达阈值，或还没跑 fulltext 步骤）","pmcid":null,"license":null,"chars":0,"source":null,"fetched_at":null}}
